Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3158G>A (p.Arg1053His), citing Ambry Variant Classification Scheme 2023: The p.R1007H variant (also known as c.3020G>A), located in coding exon 27 of the KIF1B gene, results from a G to A substitution at nucleotide position 3020. The arginine at codon 1007 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
