Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001458.5(FLNC):c.301T>C (p.Ser101Pro), citing Ambry Variant Classification Scheme 2023: The p.S101P variant (also known as c.301T>C), located in coding exon 1 of the FLNC gene, results from a T to C substitution at nucleotide position 301. The serine at codon 101 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
