Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_002769.5(PRSS1):c.300C>G (p.Asp100Glu), citing Ambry Variant Classification Scheme 2023: The p.D100E variant (also known as c.300C>G), located in coding exon 3 of the PRSS1 gene, results from a C to G substitution at nucleotide position 300. The aspartic acid at codon 100 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
