Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000020.3(ACVRL1):c.299C>G (p.Ser100Cys), citing Ambry Variant Classification Scheme 2023: The p.S100C variant (also known as c.299C>G), located in coding exon 2 of the ACVRL1 gene, results from a C to G substitution at nucleotide position 299. The serine at codon 100 is replaced by cysteine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6500 samples (13000 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be probably damaging by PolyPhen but tolerated by SIFT in silico analyses. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
