Uncertain significance — the classification assigned by Ambry Genetics to NM_006904.7(PRKDC):c.2999A>T (p.Lys1000Ile), citing Ambry Variant Classification Scheme 2023: The p.K1000I variant (also known as c.2999A>T), located in coding exon 26 of the PRKDC gene, results from an A to T substitution at nucleotide position 2999. The lysine at codon 1000 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
