NM_000271.5(NPC1):c.2998C>T (p.Pro1000Ser) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P1000S variant (also known as c.2998C>T), located in coding exon 20 of the NPC1 gene, results from a C to T substitution at nucleotide position 2998. The proline at codon 1000 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
