Uncertain significance — the classification assigned by Ambry Genetics to NM_003628.6(PKP4):c.2967T>A (p.Asn989Lys), citing Ambry Variant Classification Scheme 2023: The p.N989K variant (also known as c.2967T>A), located in coding exon 17 of the PKP4 gene, results from a T to A substitution at nucleotide position 2967. The asparagine at codon 989 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
