Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.3102A>G (p.Ile1034Met), citing Ambry Variant Classification Scheme 2023: The p.I988M variant (also known as c.2964A>G), located in coding exon 26 of the KIF1B gene, results from an A to G substitution at nucleotide position 2964. The isoleucine at codon 988 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
