Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.295C>A (p.Leu99Ile), citing Ambry Variant Classification Scheme 2023: The p.L99I variant (also known as c.295C>A), located in coding exon 1 of the CHEK2 gene, results from a C to A substitution at nucleotide position 295. The leucine at codon 99 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
