NM_001148.6(ANK2):c.11229A>T (p.Gln3743His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q3743H variant (also known as c.11229A>T), located in coding exon 42 of the ANK2 gene, results from an A to T substitution at nucleotide position 11229. The glutamine at codon 3743 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
