NM_001184.4(ATR):c.2929G>C (p.Glu977Gln) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E977Q variant (also known as c.2929G>C), located in coding exon 14 of the ATR gene, results from a G to C substitution at nucleotide position 2929. The glutamic acid at codon 977 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
