Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.2925T>A (p.Asp975Glu), citing Ambry Variant Classification Scheme 2023: The p.D975E variant (also known as c.2925T>A), located in coding exon 21 of the KIT gene, results from a T to A substitution at nucleotide position 2925. The aspartic acid at codon 975 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
