Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.2909A>G (p.Asp970Gly), citing Ambry Variant Classification Scheme 2023: The p.D970G variant (also known as c.2909A>G), located in coding exon 19 of the FLNA gene, results from an A to G substitution at nucleotide position 2909. The aspartic acid at codon 970 is replaced by glycine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6134 samples with coverage at this position. This amino acid position is completely conserved on sequence alignment. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
