Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000077.5(CDKN2A):c.28G>A (p.Glu10Lys), citing Ambry Variant Classification Scheme 2023: The p.E10K variant (also known as c.28G>A), located in coding exon 1 of the CDKN2A gene, results from a G to A substitution at nucleotide position 28. The glutamic acid at codon 10 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
