NM_001148.6(ANK2):c.2891T>C (p.Ile964Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I964T variant (also known as c.2891T>C), located in coding exon 26 of the ANK2 gene, results from a T to C substitution at nucleotide position 2891. The isoleucine at codon 964 is replaced by threonine, an amino acid with similar properties. A different variant affecting this codon (p.I964V, c.2890A>G) was detected in an individual from a cohort with prolonged QTc interval who may have had a non-genetic explanation for QTc prolongation, and co-occurred with a DSP variant in an arrhythmogenic right ventricular cardiomyopathy cohort (Gibbs C et al. J Am Heart Assoc, 2018 Aug;7:e009706; Roberts JD et al. J. Clin. Invest., 2019 07;129:3171-3184). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
