NM_001365951.3(KIF1B):c.2994G>C (p.Glu998Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E952D variant (also known as c.2856G>C), located in coding exon 25 of the KIF1B gene, results from a G to C substitution at nucleotide position 2856. The glutamic acid at codon 952 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
