Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_021076.4(NEFH):c.2852C>T (p.Ala951Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: NEFH c.2852C>T (p.Ala951Val) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.5e-05 in 228304 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2852C>T in individuals affected with Charcot-Marie Tooth Disease, Axonal, Type 2CC and no experimental evidence demonstrating its impact on protein function have been reported.
