This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 278, where T is replaced by A; at the protein level this means replaces valine at residue 93 with aspartic acid — a missense variant. Submitter rationale: The p.V93D variant (also known as c.278T>A), located in coding exon 4 of the CFTR gene, results from a T to A substitution at nucleotide position 278. The valine at codon 93 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is conserved in all available species, except in fish. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
