Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.2779G>A (p.Val927Ile), citing Ambry Variant Classification Scheme 2023. This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 2779, where G is replaced by A; at the protein level this means replaces valine at residue 927 with isoleucine — a missense variant. Submitter rationale: The p.V927I variant (also known as c.2779G>A), located in coding exon 23 of the FBN1 gene, results from a G to A substitution at nucleotide position 2779. The valine at codon 927 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
