NM_000249.4(MLH1):c.1009T>C (p.Ser337Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S337P variant (also known as c.1009T>C), located in coding exon 11 of the MLH1 gene, results from a T to C substitution at nucleotide position 1009. The serine at codon 337 is replaced by proline, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species.
