Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.2722A>C (p.Asn908His), citing Ambry Variant Classification Scheme 2023: The p.N908H variant (also known as c.2722A>C), located in coding exon 18 of the DNAH5 gene, results from an A to C substitution at nucleotide position 2722. The asparagine at codon 908 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
