Uncertain significance — the classification assigned by Ambry Genetics to NM_024642.5(GALNT12):c.270G>C (p.Glu90Asp), citing Ambry Variant Classification Scheme 2023: The p.E90D variant (also known as c.270G>C), located in coding exon 1 of the GALNT12 gene, results from a G to C substitution at nucleotide position 270. The glutamic acid at codon 90 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
