Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.270dup (p.Lys91Ter), citing Ambry Variant Classification Scheme 2023: The c.270dupT pathogenic mutation, located in coding exon 4 of the PMS2 gene, results from a duplication of T at nucleotide position 270, causing a translational frameshift with a predicted alternate stop codon (p.K91*). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
