Uncertain significance for Nephrolithiasis/nephrocalcinosis — the classification assigned by Ambry Genetics to NM_000388.4(CASR):c.2704C>T (p.Leu902Phe), citing Ambry Variant Classification Scheme 2023: The p.L902F variant (also known as c.2704C>T), located in coding exon 6 of the CASR gene, results from a C to T substitution at nucleotide position 2704. The leucine at codon 902 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
