Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.110C>T (p.Thr37Ile), citing Ambry Variant Classification Scheme 2023: The p.T37I variant (also known as c.110C>T), located in coding exon 2 of the PMS2 gene, results from a C to T substitution at nucleotide position 110. The threonine at codon 37 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
