Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.2803G>C (p.Asp935His), citing Ambry Variant Classification Scheme 2023: The p.D889H variant (also known as c.2665G>C), located in coding exon 24 of the KIF1B gene, results from a G to C substitution at nucleotide position 2665. The aspartic acid at codon 889 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
