NM_004415.4(DSP):c.5757T>G (p.Asp1919Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1919E variant (also known as c.5757T>G), located in coding exon 24 of the DSP gene, results from a T to G substitution at nucleotide position 5757. The aspartic acid at codon 1919 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
