NM_001232.4(CASQ2):c.265A>G (p.Ile89Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I89V variant (also known as c.265A>G), located in coding exon 2 of the CASQ2 gene, results from an A to G substitution at nucleotide position 265. The isoleucine at codon 89 is replaced by valine, an amino acid with highly similar properties. This variant was reported as heterozygous in an exome cohort; however, clinical details were not provided (Landstrom AP et al. Circ Arrhythm Electrophysiol, 2017 Apr;10:[Epub ahead of print]). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
