Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.264A>C (p.Glu88Asp), citing Ambry General Variant Classification Scheme_2022: The p.E88D variant (also known as c.264A>C), located in coding exon 1 of the PALLD gene, results from an A to C substitution at nucleotide position 264. The glutamic acid at codon 88 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
