Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.2621G>A (p.Cys874Tyr), citing Ambry Variant Classification Scheme 2023: The p.C874Y variant (also known as c.2621G>A), located in coding exon 17 of the TRPM4 gene, results from a G to A substitution at nucleotide position 2621. The cysteine at codon 874 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
