Pathogenic for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.2605dup (p.Ile869fs), citing Ambry Variant Classification Scheme 2023: The c.2605dupA pathogenic mutation, located in coding exon 15 of the CFTR gene, results from a duplication of A at nucleotide position 2605, causing a translational frameshift with a predicted alternate stop codon (p.I869Nfs*27). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
