Uncertain significance — the classification assigned by Ambry Genetics to NM_004725.4(BUB3):c.25C>A (p.Leu9Met), citing Ambry Variant Classification Scheme 2023: The p.L9M variant (also known as c.25C>A), located in coding exon 1 of the BUB3 gene, results from a C to A substitution at nucleotide position 25. The leucine at codon 9 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
