NM_001430.5(EPAS1):c.2596C>A (p.Leu866Met) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L866M variant (also known as c.2596C>A), located in coding exon 16 of the EPAS1 gene, results from a C to A substitution at nucleotide position 2596. The leucine at codon 866 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
