Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001232.4(CASQ2):c.258T>G (p.His86Gln), citing Ambry Variant Classification Scheme 2023: The p.H86Q variant (also known as c.258T>G), located in coding exon 2 of the CASQ2 gene, results from a T to G substitution at nucleotide position 258. The histidine at codon 86 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
