Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001365276.2(TNXB):c.2585G>A (p.Arg862His), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the TNXB gene (transcript NM_001365276.2) at coding-DNA position 2585, where G is replaced by A; at the protein level this means replaces arginine at residue 862 with histidine — a missense variant. Submitter rationale: Variant summary: TNXB c.2585G>A (p.Arg862His) results in a non-conservative amino acid change located in the Fibronectin type III domain (IPR003961) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 245956 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2585G>A in individuals affected with Ehlers-Danlos syndrome due to tenascin-X deficiency and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1793424). Based on the evidence outlined above, the variant was classified as uncertain significance.
