Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.2573T>A (p.Val858Asp), citing Ambry Variant Classification Scheme 2023: The p.V858D variant (also known as c.2573T>A), located in coding exon 6 of the PALB2 gene, results from a T to A substitution at nucleotide position 2573. The valine at codon 858 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
