Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004006.3(DMD):c.2573C>T (p.Thr858Ile), citing Ambry Variant Classification Scheme 2023: The p.T858I variant (also known as c.2573C>T), located in coding exon 20 of the DMD gene, results from a C to T substitution at nucleotide position 2573. The threonine at codon 858 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
