Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.254T>G (p.Met85Arg), citing Ambry Variant Classification Scheme 2023: The p.M85R variant (also known as c.254T>G), located in coding exon 2 of the TMEM127 gene, results from a T to G substitution at nucleotide position 254. The methionine at codon 85 is replaced by arginine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
