NM_017802.4(DNAAF5):c.2522G>A (p.Cys841Tyr) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C841Y variant (also known as c.2522G>A), located in coding exon 13 of the DNAAF5 gene, results from a G to A substitution at nucleotide position 2522. The cysteine at codon 841 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
