Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001165963.4(SCN1A):c.2508C>A (p.Asp836Glu), citing Ambry Variant Classification Scheme 2023: The p.D836E variant (also known as c.2508C>A), located in coding exon 14 of the SCN1A gene, results from a C to A substitution at nucleotide position 2508. The aspartic acid at codon 836 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
