NM_005751.5(AKAP9):c.2506T>C (p.Cys836Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C836R variant (also known as c.2506T>C), located in coding exon 8 of the AKAP9 gene, results from a T to C substitution at nucleotide position 2506. The cysteine at codon 836 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
