Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000268.4(NF2):c.1100C>G (p.Ala367Gly), citing Ambry Variant Classification Scheme 2023: The p.A367G variant (also known as c.1100C>G), located in coding exon 11 of the NF2 gene, results from a C to G substitution at nucleotide position 1100. The alanine at codon 367 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
