Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.2503G>T (p.Asp835Tyr), citing Ambry Variant Classification Scheme 2023: The p.D835Y variant (also known as c.2503G>T), located in coding exon 15 of the CFTR gene, results from a G to T substitution at nucleotide position 2503. The aspartic acid at codon 835 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
