NM_017950.4(CCDC40):c.2499G>C (p.Met833Ile) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M833I variant (also known as c.2499G>C), located in coding exon 15 of the CCDC40 gene, results from a G to C substitution at nucleotide position 2499. The methionine at codon 833 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
