NM_006767.4(LZTR1):c.2499C>G (p.Cys833Trp) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C833W variant (also known as c.2499C>G), located in coding exon 21 of the LZTR1 gene, results from a C to G substitution at nucleotide position 2499. The cysteine at codon 833 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
