Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.52031A>G (p.His17344Arg), citing Ambry Variant Classification Scheme 2023: The p.H8279R variant (also known as c.24836A>G), located in coding exon 100 of the TTN gene, results from an A to G substitution at nucleotide position 24836. The histidine at codon 8279 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
