NM_005751.5(AKAP9):c.2473C>T (p.Leu825Phe) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L825F variant (also known as c.2473C>T), located in coding exon 8 of the AKAP9 gene, results from a C to T substitution at nucleotide position 2473. The leucine at codon 825 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
