Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.2472T>G (p.Ile824Met), citing Ambry Variant Classification Scheme 2023: The p.I824M variant (also known as c.2472T>G), located in coding exon 14 of the CFTR gene, results from a T to G substitution at nucleotide position 2472. The isoleucine at codon 824 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
