NM_000219.6(KCNE1):c.247_248delinsAC (p.Glu83Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.247_248delGAinsAC variant (also known as p.E83T), located in coding exon 1 of the KCNE1 gene, results from an in-frame deletion of GA and insertion of AC at nucleotide positions 247 to 248. The glutamic acid at codon 83 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
