NM_001903.5(CTNNA1):c.2462C>T (p.Ala821Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A821V variant (also known as c.2462C>T), located in coding exon 17 of the CTNNA1 gene, results from a C to T substitution at nucleotide position 2462. The alanine at codon 821 is replaced by valine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
